Differential Expression of LEF1 Isoforms in Adult Lymphoid and Myeloid Malignancies

Objective: Lymphoid enhancer-binding factor-1 (LEF1) is one of the key regulators lymphocyte proliferation and its aberrant expression a prognostic factor for lymphoid or myeloid malignancies. In this study, we focused on LEF1 isoforms in several hematological malignancies found tissue-specific differential full-length (FL)-LEF1 gene tumor suppressor (∆LEF1) variant. Material Method: Fifty-three leukemia/lymphoma patients were included study. Diagnostic samples “lymphoid group” patients: Chronic Lymphoblastic Leukemia (CLL) (n=10), B-cell Acute (B-ALL) (n=9) “myeloid Myeloblastic (CML) (n=12), Myeloid (AML) (n=13), Multiple Myeloma (MM) studied. Healthy bone marrow, peripheral blood cells, CD34 positive cells used as controls. Total (T) FL-LEF1 transcript levels examined by using quantitative real-time polymerase chain reaction (qRT-PCR). T mRNA ratios also evaluated calculation ∆LEF1. Results: significantly high malignancies, but MM AML have decreased levels. Although CLL levels, ratio T/FL was decreased. Conclusion: lymphocytes not only overexpression seem to accompany leukemia progress.
